Mobile CRISPR-Cas9 based anti-phage system in E. coli

摘要: Escherichia coli is one of the most important microbial cell factories, but infection by bacteriophages in the environment may have a huge impact on its application in industrial production. Here, we developed a mobile CRISPR-Cas9 based anti-phage system for bacteriophages defense in E. coli. Two conjugative plasmids pGM1 (phosphoglucomutase 1) and pGM2 carrying one and two guide RNAs, respectively, were designed to defend against a filamentous phage. The results showed that the pGM1 and pGM2 could decrease the phage infection rate to 1.6% and 0.2% respectively in infected cells. For preventing phage infection in E. coli, the pGM2 decreased the phage infection rate to 0.1%, while pGM1 failed to block phage infection. Sequence verification revealed that point mutations in protospacer or protospacer adjacent motif sequences of the phage genome caused loss of the defense function. These results support the potential application of MCBAS in E. coli cell factories to defend against phage infections.

摘要: 通过筛选不同的碱基编辑器,发现广泛使用的酿脓链球菌CRISPR相关蛋白9(SpCas9)或毛螺科菌Cpf1(LbCpf1)胞嘧啶碱基编辑器在分枝杆菌中不具有活性,而嗜热链球菌Cas9(St1Cas9)胞嘧啶碱基编辑器活性较好虽然使用St1Cas9 胞嘧啶碱基编辑器能够实现C到T的碱基编辑,但是,在碱基编辑过程中却产生了大量的副产物。将尿嘧啶-N-糖基化酶抑制剂或尿嘧啶-N-糖基化酶分别融合到St1Cas9 胞嘧啶碱基编辑器中,得到了两种新的碱基编辑器——CTBE和CGBE此外,由于野生型St1Cas9 在靶向DNA序列时需要识别严格的前间隔序列邻近基序(PAM),因此,通过结构介导的蛋白质工程设计了PAM扩展的St1Cas9,扩大了碱基编辑器的靶向范围。

具有四方ThMn12型结构的富铁化合物有潜力满足当下人们对于高磁能积、工作温度为150~200 ℃的贫稀土永磁体的需求。尽管磁体制备技术的发展滞后于相关磁性材料的研究是正常的,但对于ThMn12型磁性材料而言,这种研究进展的差异非常显著。人们发现,引入镧(La)可以提高Sm(Fe,Ti)12中少数低熔点相的稳定性,从而首次使得液相烧结成为可能。然而,La金属的高反应活性会明显地破坏材料的矫顽力(Hc)。,Sm)(Fe,Ti)12合金中也可以实现。机械化学合成的(Sm,Zr)(Fe,Si)12和(Sm,Zr)(Fe,Co,Ti)12粉末可能适合烧结成具有高强度的全致密磁体,尽管在此之前,两者都已开发出较高的各向异性

CRISPR-Cas9 mediated LAG-3 disruption in CAR-T cells

摘要:

T cells engineered with chimeric antigen receptor (CAR) have been successfully applied to treat advanced refractory B cell malignancy. However, many challenges remain in extending its application toward the treatment of solid tumors. The immunosuppressive nature of tumor microenvironment is considered one of the key factors limiting CAR-T efficacy. One negative regulator of T cell activity is lymphocyte activation gene-3 (LAG-3). We successfully generated LAG-3 knockout T and CAR-T cells with high efficiency using CRISPR-Cas9 mediated gene editing and found that the viability and immune phenotype were not dramatically changed during in vitro culture. LAG-3 knockout CAR-T cells displayed robust antigen-specific antitumor activity in cell culture and in murine xenograft model, which is comparable to standard CAR-T cells. Our study demonstrates an efficient approach to silence immune checkpoint in CAR-T cells via gene editing.

Generation of CRISPR/Cas9-mediated lactoferrin-targeted mice by pronuclear injection of plasmid pX330

摘要: Lactoferrin is a member of the transferrin family of multifunctional iron binding glycoproteins. While numerous physiological functions have been described for lactoferrin, the mechanisms underlying these functions are not clear. To further study the functions and mechanisms of lactoferrin, we modified the lactoferrin promoter of mice using the CRISPR/Cas9 system to reduce or eliminate lactoferrin expression. Seven mice with lactoferrin promoter mutations were obtained with an efficiency of 24% (7/29) by injecting the plasmid pX330, expressing a small guide RNA and human codon-optimized SpCas9, into fertilized eggs of mice. Plasmid integration and off-targeting of pX330 were not detected. These results confirmed that pronuclear injection of a circular plasmid is a feasible and efficient method for targeted mutagenesis in mice.

Growth suppression of colorectal cancer expressing S492R EGFR by monoclonal antibody CH12

Colorectal cancer (CRC) is a common malignant tumor in the digestive tract, and 30%–85% of CRCs express epidermal growth factor receptors (EGFRs). Recently, treatments using cetuximab, also named C225, an anti-EGFR monoclonal antibody, for CRC have been demonstrated to cause an S492R mutation in EGFR. However, little is known about the biological function of S492R EGFR. Therefore, we attempted to elucidate its biological function in CRC cells and explore new treatment strategies for this mutant form. Our study indicated that EGFR and S492R EGFR accelerate the growth of CRC cells in vitro and in vivo and monoclonal antibody CH12, which specifically recognizes an EGFR tumor-specific epitope, can bind efficiently to S492R EGFR. Furthermore, mAb CH12 showed significantly stronger growth suppression activities and induced a more potent antibody-dependent cellular cytotoxicity effect on CRC cells bearing S492R EGFR than mAb C225. mAb CH12 obviously suppressed the growth of CRC xenografts with S492R EGFR mutations in vivo. Thus, mAb CH12 may be a promising therapeutic agent in treating patients with CRC bearing an S492R EGFR mutation.

摘要: 提出一种由时滞线性智能体组成的多智能体系统的领导-跟随同步算法。各智能体中存在的不同时滞会导致不收敛于零的同步误差。但是,可限制误差范数并给出误差边界。利用线性矩阵不等式对主要结果进行验证,且该问题的规模与智能体数量无关。通过案例对结果进行说明,强调稳定误差是由异构延迟引起的事实,并验证该算法在一定误差范围内有实现同步的能力。

Facile controlled synthesis of hierarchically structured mesoporous Li4Ti5O12/C/rGO composites as high-performance

摘要: In this paper, a facile strategy is proposed to controllably synthesize mesoporous Li4Ti5O12/C nanocomposite embedded in graphene matrix as lithium-ion battery anode via the co-assembly of Li4Ti5O12 (LTO) precursor, GO, and phenolic resin. The obtained composites, which consists of a LTO core, a phenolic-resin-based carbon shell, and a porous frame constructed by rGO, can be denoted as LTO/C/rGO and presents a hierarchical structure. Owing to the advantages of the hierarchical structure, including a high surface area and a high electric conductivity, the mesoporous LTO/C/rGO composite exhibits a greatly improved rate capability as the anode material in contrast to the conventional LTO electrode.

Poor adherence to P2Y12 antagonists increased cardiovascular risks in Chinese PCI-treated patients

Low adherence to secondary prevention medications (ATM) of patients after acute coronary syndrome (ACS) is associated with poor clinical outcomes. However, literature provides limited data on assessment of ATM and risks associated with poor in Chinese patients with ACS. In the current work, ATM was assessed in consecutively recruited patients with ACS in Tongji Hospital from November 5, 2013 to December 31, 2014. A total of 2126 patients were classified under low adherence (proportion of days covered (PDC)<50%) and high adherence (PDC>50%) groups based on their performance after discharge. All patients were followed up at the 1st, 6th, and 12th month of discharge while recording ATM and major adverse cardiac events (MACE). Bivariate logistic regression was used to identify the factors associated with ATM. Cox regression was used to analyze the association between ATM and MACE within one year after discharge. Results showed that coronary artery bypass grafting (CABG) alone had significantly lower proportion of high adherence to P2Y12 antagonists (83.0% vs. 90.7%, P<0.01) than patients treated with percutaneous coronary intervention (PCI) only. Moreover, in patients undergoing PCI, high adherence to P2Y12 antagonists decreased the risk of MACE (hazard ratio=0.172, 95% confidence interval: 0.039–0.763; P=0.021). In conclusion, PCI-treated patients are more prone to remaining adherent to medications than CABG-treated patients. High adherence to P2Y12 antagonists was associated with lower risk of MACE.
